Background
Scarab’s Clean Genome® strains were specifically designed for the production of biotherapeutic protein and DNA. The “cleanest” medium to use for biotherapeutic production is a chemically defined, minimal medium. Accordingly, Ultra HD Minimal Batch Media formulation resulted from Scarab Genomics’ systematic determination of the metabolic needs of the Clean Genome® E. coli strains in fed batch fermentation for a variety of protein and DNA targets. Ultra HD Minimal Batch Media together with the optimized metabolism of the Clean Genome® strains enable ultra high densities up to 300 OD600 resulting in proteins yields of >40 g/L of test protein and 700 – 1000 mg/L plasmid in 10L scale fermentations without The medium consists of phosphate buffer, magnesium, ferric citrate, trace elements, and uses glucose as the carbon source. The same base medium used for the batch phase of fed-batch fermentation can be used in optimizing expression in shake flasks, thereby providing continuity between the two processes. A separate Feed Medium (Cat. No. S300-F-1L) supplies glucose, magnesium, iron, and trace elements for the feeding stage of fed-batch fermentation.
